Ciliary Regulation: Disassembly Takes the Spotlight

چکیده
The mechanisms that control organelle size are poorly understood. Genetic analysis now shows that a NIMA-related kinase helps to regulate the size of cilia by activating disassembly of the cilium when it gets too long.

Deacetylation of α-tubulin and cortactin is required for HDAC6 to trigger ciliary disassembly.
Cilia play important roles in sensing extracellular signals and directing fluid flow. Ciliary dysfunction is associated with a variety of diseases known as ciliopathies. Histone deacetylase 6 (HDAC6) has recently emerged as a major driver of ciliary disassembly, but little is known about the downstream players. Here we provide the first evidence that HDAC6-mediated deacetylation of α-tubulin an...
متن کاملPCM1 recruits Plk1 to the pericentriolar matrix to promote primary cilia disassembly before mitotic entry.
Primary cilia, which emanate from the cell surface, exhibit assembly and disassembly dynamics along the progression of the cell cycle. However, the mechanism that links ciliary dynamics and cell cycle regulation remains elusive. In the present study, we report that Polo-like kinase 1 (Plk1), one of the key cell cycle regulators, which regulate centrosome maturation, bipolar spindle assembly and...
متن کاملThe Primary Cilium: Keeper of the Key to Cell Division
Assembly of the nonmotile primary cilium of vertebrate cells requires one of the centrioles of the centrosome. A cluster of new studies, including one in this issue of Cell by Pugacheva et al. (2007), reveal that ciliary assembly proteins influence cell-cycle progression and that a centrosomal "mitotic kinase" promotes ciliary disassembly. The link between the cell cycle and the primary cilium ...
متن کاملThe nephronophthisis gene product NPHP2/Inversin interacts with Aurora A and interferes with HDAC6-mediated cilia disassembly.
BACKGROUND Nephronophthisis (NPH) is a rare recessive disease caused by several different gene mutations. Most gene products localize to the cilium, and thus, the various NPH manifestations including kidney cysts and situs inversus have been linked to ciliary defects. RESULTS Here, we describe that targeted knockdown of NPHP2 significantly reduced the number of cilia on polarized MDCK cells. ...
متن کاملThe Kinases LF4 and CNK2 Control Ciliary Length by Feedback Regulation of Assembly and Disassembly Rates
BACKGROUND Many of the diverse functions of cilia depend upon tight control of their length. Steady-state length reflects a balance between rates of ciliary assembly and disassembly, two parameters likely controlled by a length sensor of unknown identity or mechanism. RESULTS A null mutation in Chlamydomonas CNK2, a member of the evolutionarily conserved family of NIMA-related kinases, reveal...
